Fluorescence properties of doxorubicin in PBS buffer and PVA films.

Fluorescence properties of doxorubicin in PBS buffer and PVA films.

J Photochem Photobiol B. 2017 Mar 30;170:65-69

Abstract
We studied steady-state and time-resolved fluorescence properties of an anticancer drug Doxorubicin in a saline buffer and poly-vinyl alcohol (PVA) film. Absorption of Doxorubicin, located at blue-green spectral region, allows a convenient excitation with visible light emitting diodes or laser diodes. Emission of Doxorubicin with maximum near 600nm can be easily detected with photomultipliers and CCD cameras. Both, absorption and fluorescence spectra in polymeric matrix show more pronounced vibronic structures than in solution. Also, the steady-state anisotropy in the polymer film is significantly higher than in the saline solution. In PVA film the fluorescence anisotropy is about 0.30 whereas in the saline buffer only 0.07. Quantum efficiencies of Doxorubicin were compared to a known standard Rhodamine 101 which has fluorescence emission in a similar spectral region. The quantum yield of Doxorubicin in PVA film is more than 10% and about twice higher than in the saline solution. Similarly, the lifetime of doxorubicin in PVA film is about 2ns whereas in the saline solution only about 1ns. The fluorescence anisotropy decays show that Doxorubicin molecules are freely rotating in the saline buffer with a correlation time of about 290ps, and are almost completely immobilized in the PVA film. The spectroscopic investigations presented in this manuscript are important, as they provide answers to changes in molecular properties of Doxorubicin depending changes in the local environment, which is useful when synthesizing nanoparticles for Doxorubicin entrapment.

A green analytical chemistry approach for lipid extraction: computation methods in the selection of green solvents as alternative to hexane.

Abstract
There is a great interest in finding alternatives and green solvents in extraction processes to replace petroleum based solvents. In order to investigate these possibilities, computational methods, as Hansen solubility parameters (HSP) and conductor-like screening model for real solvent (COSMO-RS), were used in this work to predict the solvation power of a series of solvents in salmon fish lipids. Additionally, experimental studies were used to evaluate the performance in lipids extraction using 2-methyltetrahydrofurane, cyclopentyl methyl ether, dimethyl carbonate, isopropanol, ethanol, ethyl acetate, p-cymene and d-limonene compared with hexane. Lipid classes of extracts were obtained by using high performance thin-layer chromatography (HPTLC), whereas gas chromatography with a flame ionization detector (GC/FID) technique was employed to obtain fatty acid profiles. Some differences between theoretical and experimental results were observed, especially regarding the behavior of p-cymene and d-limonene, which separate from the predicted capability. Results obtained from HPTLC indicated that p-cymene and d-limonene extract triglycerides (TAGs) and diglycerides (DAGs) at levels of 73 and 19%, respectively, whereas the other studied extracts contain between 75 and 76% of TAGs and between 16 and 17% of DAGs. Fatty acid profiles, obtained by using GC-FID, indicated that saturated fatty acids (SFAs) between 19.5 and 19.9% of extracted oil, monounsaturated fatty acids (MUFAs) in the range between 43.5 and 44.9%, and PUFAs between 31.2 and 34.6% were extracted. p-Cymene and limonene extracts contained lower percentages than the other studied solvents of some PUFAs due probably to the fact that these unsaturated fatty acids are more susceptible to oxidative degradation than MUFAs. Ethyl acetate has been found to be the best alternative solvent to hexane for the extraction of salmon oil lipids. Graphical Abstract ᅟ.

Abstract
Dengue and chikungunya are arboviral diseases mainly vectored by the mosquito Aedes aegypti. Presently, there is no treatment for these viral diseases and their prevention is still based on vector control measures. Nanopesticides fabricated using herbal extracts as reducing and capping agents currently represent an excellent platform for pest control. In this scenario, the present study assessed the acute toxicity of seven plants employed in ethno-veterinary medicine of southern India, as well as the green synthesis of zinc oxide nanoparticles, on third-instar larvae of A. aegypti. Larvae were exposed to extracts of the seven plants obtained with solvents of different polarity (acetone, ethanol, petroleum ether, and water) for 24 h. Maximum efficacy was observed for Lobelia leschenaultiana leaf extracts prepared using all the four solvent extracts (LC50 = 22.83, 28.12, 32.61, and 36.85 mg/L, respectively). Therefore, this plant species was used for the synthesis and stabilization of ZnO nanoparticles based on its maximum efficacy against third-instar larvae of A. aegypti. L. leschenaultiana-encapsulated ZnO nanoparticles showed 100% mortality when tested at 10 mg/L, the LC50 was extremely low,  1.57 mg/L. Zinc acetate achieved only 65.33% when tested at 60 mg/L, with a LC50 of 51.62 mg/L. Additionally, ZnO nanoparticles inhibited growth of Pseudomonas aeruginosa, Proteus vulgaris, Shigella sonnei, and Vibrio parahaemolyticus and also inhibited biofilm formation on selected microbila pathogens, showing impact on EPS production and hydrophobicity. Overall, our results suggest that L. leschenaultiana-fabricated ZnO nanoparticles have a significant potential to control A. aegypti mosquitoes and Gram-negative bacterial pathogens.

Nicotine dependence is associated with depression and childhood trauma in smokers with schizophrenia: results from the FACE-SZ dataset.

Abstract
In a perspective of personalized care for smoking cessation, a better clinical characterization of smokers with schizophrenia (SZ) is needed. The objective of this study was to determine the clinical characteristics of SZ smokers with severe nicotine (NIC) dependence. 240 stabilized community-dwelling SZ smokers (mean age = 31.9 years, 80.4% male gender) were consecutively included in the network of the FondaMental Expert Centers for Schizophrenia and assessed with validated scales. Severe NIC dependence was defined by a Fagerstrom questionnaire score ≥ 7. Depression was defined by a Calgary score ≥ 6. Childhood trauma was self-reported by the Childhood Trauma Questionnaire score (CTQ). Ongoing psychotropic treatment was recorded. Severe NIC dependence was identified in 83 subjects (34.6%), depression in 60 (26.3%). 44 (22.3%) subjects were treated by antidepressants. In a multivariate model, severe NIC dependence remained associated with depression (OR = 3.2, p = 0.006), male gender (OR = 4.5, p = 0.009) and more slightly with childhood trauma (OR = 1.03, p = 0.044), independently of socio-demographic characteristics, psychotic symptoms severity, psychotropic treatments and alcohol disorder. NIC dependence was independently and strongly associated with, respectively, depression and male gender in schizophrenia, and only slightly with history of childhood trauma. Based on these results, the care of both nicotine dependence and depression should be evaluated for an effective smoking cessation intervention in schizophrenia.
